Zen always collects a Z-stack from
the bottom to the top
Note:
-- Total Z-series collection time is: "Scan time" for one focus position multiplied by the
number of "slices" (focus points). 30-45 minutes collection times are common.
-- To best match the manual live focus position always fopcus from coverslip side to
slide side (i.e. positive to negative). If you go past go back down and approach
again.
-- Optimal section step is half the Airy section thick. This is to satisfy the the Nyquist
sampling criteria
-- Transmitted images are NOT confocal. Do not collect T-PMT in Z-series.
Z-SERIES CAPTURE
1. Find and focus your sample and set capture
parameters as normal.
2. Select Z-STACK in configuration setup area.
3. Open focus tools. SHOW ALL selected. Click ZERO
POSITION ö MANUALLY. to reset current focus position
as "Zero" position.
4. Confirm clicked.

5. Use Live view. Focus past the top of your sample,
come back to the top, and Stop live. [If using sequential
scanning, you only need one channel to set focus]
6. Open Z-STACK TOOLS. Check Position if the position
is a positive number click "Set First".
[Positive is towards the slide, negative to towards the
coverslip]
7. Focus to the bottom of your sample. In Z-STACK
TOOLS: Check Position if the position number is
negative. click "Set Last".
8. Click the < OPTIMAL> setting button to set interval (stepsize)
9. Check "brightness" of the middle of your sample (the brightest region), make sure you will not
over-saturate when collecting the images here.
10. Click "Start Experiment" to collect the Z-series
