ZEN SOFTWARE

OCULAR:

—> Software control of Microscope stand. Select OCULAR tab to view through eye-pieces

1. OCULAR >> ONLINE

2. <Click> MICROSCOPE START button

3. LOCATE SAMPLE with normal microscopy

(BF, DIC or Epi-Fluorescence)

ACQUISITION:

—> Software control for Confocal Imaging.

–-> LOCATE SAMPLE with normal microscopy, first.

1. DYE SETUP:

a. open SMART SETUP menu

b. <click> on Triangle under DYE

c. either:

select Dye from RECENT dyes list

or in SEARCH type name of the desired dye and select

from sorted DYES list

d. repeat steps b & c until all dyes are listed

e. select FASTEST for simultaneous collection

BEST SIGNAL for sequential collection [See Sequential

Setup]

f. <click> - APPLY

2. ACQUISITION MODE menu:

–> Check T Show all box

a. FRAME SIZE to 1024 X 1024

b. SPEED to 5 [ lower number = slower scan = better Signal-

to-Noise ratio]

c. AVERAGING: set NUMBER to 1

d. SCAN AREA: <click> - RESET ALL

3. CHANNELS

–> Check T Show all box

a. For Each dye: Select CHANNEL

PINHOLE –> 1AU

GAIN (MASTER): Set PMT to “Normal” value for your sample

(400-700 typically)

DIGITAL OFFSET to 0%

ö Set OFFSET to 0% (0-10 is acceptable)

DIGITAL GAIN to 1.0x

Remember to Reset

PINHOLE to 1 AU

Hint: The software is very complex but tries to help with “context sensitive” mouse commands,

“Right-Clicking” items on the screen will often show you commands for what you are

looking for.

— Many items can be dragged and dropped, tools can be re-sized and floated.
