The shutters can be controlled either in the software or on the microscope
Brightfield:
- Select channel 1 in Volocity (this will open the shutter)
- Select the type of contrast (Brightfield / Phase Contrast / DIC) on the condenser
- Adjust the brightness using the rocker switch on the front of the microscope
- The shutter can be closed either in the software or on the filter wheel pad (S1)
Fluorescence:
- Select fluorescent channel in Volocity (this will open the shutter)
- The shutter can be closed either in the software or on the microscope (FL button – right-hand side)
Adjusting brightfield Köhler illumination
• Always recommended, essential for brightfield image
• Switch to 10x objective
• Open brightfield shutter with S1 button on Keypad
• Adjust brightness with large black wheel on front / bottom
of microscope)
• Make sure condenser iris is not completely closed (wheel
on side of condenser)
• Focus on your sample
• Completely close field iris (top silver lever) (if the image
turns completely black, reopen until you see some light,
proceed to next step and close again after you have
focussed the iris)
• Focus iris using black condenser focus control
• Centre the iris with the 2 silver Köhler screws
• Re-open field iris until the edge is just not visible anymore
• Re-adjust field iris every time you switch to a different objective
