Time series

• in "Acquisition Setup" select the

“Time” tab

• set intervals

• set duration

Multi-position

Stage needs to be calibrated for this (see

startup), then in the "Acquisition Setup" select

“Points” tab and set "Change XY using" to

"Ludl XY Stage"

Manual positioning

• In "Stage" menu - "Clear All Points"

• In "Video" menu set to "Live Video"

• Move to the desired point

• In "Stage" menu - "Add Point"

• In "Stage" menu - "Review Points" allows checking and re-adjusting of the focus for each of your

points separately

Automated positioning

• In "Video" set to "XY Stage

• In "Stage" - "Make Well Overlay"

• In "Stage" - "Create Points from Well Overlay"

• Follow the instructions

Stitching

Stage needs to be calibrated for this (see startup),

then in the "Acquisition Setup" select th “Stitch” tab

and set "Change XY using" to "Ludl XY Stage"

Select from ROI – draw region in preview

• In "Video" set to "XY Stage

• Draw ROI

Fixed Size

• Select tile dimensions

Options

• Overlap – 10% recommended

• Create stitched image –not recommended if

image is large or has large areas of undefined

signal

• Save raw tiles -recommended
